Does DNA testing work?

Do those DNA tests work? Do you need to already have your family tree mapped out for the DNA tests to work? We registered for ancestry.com but without pre existing knowledge it was useless. I am hoping DNA test would be easier. Who has the best reputation for accurate DNA testing if this process works at all?
